If there's something with an interesting setup, I prefer to go for that. If it's suggestive, I'll always choose a creative premise over just "character x character" or a pinup.
Ideally it would be something without too many characters, because most requests I do are simple, quick sketches.
I give preference to characters I like drawing more. Celestia for instance is difficult for me to draw while conversely I can make Snails much more expressive.
Focus on the mane cast is given precedence. I'm not very interested in drawing most of the background ponies and even lesser characters like Trixie don't get a lot of art from me, though I do try to diversify from time to time.
Lastly, humor will always win out, so if it's a clever or funny or just unique idea I'll always take that first.
