Ok, so I will take a stab at our patented /co/ brand overanalysis character building with regards to Daisy.

When we saw the flower girls run from Zecora at the end of Bridle Gossip, Rose ran into a separate house and disappeared. Lily fled into the flower shop, followed by Daisy. When Twilight came to the door, she came looking for Daisy.

Considering the flower shop is covered in Daisies, and its sign is a big Daisy, I am going to guess that Daisy owns it, and that Lily is her employee. Rose probably just lives next door, but is good friends with the two due to the whole flower thing.
