I mean, they each already have distinctive designs, visually-distinguishable from the side, vectored in Flash with fluid movement animations.
And they each have unique abilities and characteristics that would be useful in a platformer:
>Twilight Sparkle: Magic, and friendship. Not very strong. (Magic) >Rarity: Gem-finding (also dressmaking, and manipulation via whining and complaining, although this may not be as useful). Dislikes dirt. (Generosity) >Fluttershy: Good with animals; also, has The Stare™. Timid (until provoked). Can fly. (Kindness) >Rainbow Dash: Can fly very quickly and fast; rapid acceleration, but easy to "Rainbow Crash" her. (Loyalty) >Applejack: Strong, can pull things, can kick, has apples. (Honesty) >Pinkie Pie: Can break the fourth wall (so knows she's in a platformer). Moves very quickly. Can probably double-jump, definitely bounce, find balloons/flying machines/etc from nowhere. (Laughter) >Spike: Eh... I got nuthin'. Oh! Messages to Princess Celestia of course, and a wisecracking sidekick to be sarcastic at you.
Plots could be varied: spooky castle in the Everfree Forest, to rescue the Cutie Mark Crusaders who have gotten lost/trapped/captured while venturing out (on a quest to find their cutie marks, naturally). Dialogue between ponies could be fun.
